چکیده

• A rapid, reproducible method of determiniEg the presence and relative amounts of adhesive and aggregated blood elements in the blood has recently been developed in this laboratory. The method involves measuring the pressure required to force blood at a constant rate through a standardized screen with multiple openings 20 micra square and 20 micra deep. This pressure has been named "screen nitration pressure." During storage of blood and during exsanguination, platelets and leucocytes form aggregates which cause remarkable increases in this pressure. It has also been observed that adenosine diphosphate; a component of platelets, causes platelets suspended in plasma to aggregate, and that monoamine oxidase inhibitors inhibit platelet aggregations. These observations have led to an interest in the composition of the platelets, and to further studies of the effects of certain components of platelets on the suspension stability of blood. One of these constituents, 5-hydroxytryptamine (serotonin), has become particularly interesting in this respect.

Aspirin at therapeutic concentrations does not affect 5-hydroxytryptamine uptake by platelets.

Aspirin at therapeutic concentrations does not inhibit the uptake of 5-hydroxytryptamine (5-HT) by blood platelets nor induce release of 5-HT from platelets, although platelet aggregation responses to collagen and arachidonic acid (which are dependent on platelet prostaglandin synthesis) are abolished. This does not support an earlier claim that aspirin's effects on platelet function include in...

5-Hydroxytryptamine levels and platelet aggregation responses in subjects with acute migraine headache.

Blood 5-hydroxytryptamine levels were reduced during migraine attacks in patients who had not taken any drugs but the aggregation responses to 5-hydroxytryptamine of the blood platelets from these patients were similar to responses of platelets from migrainous subjects between attacks. This confirms earlier findings that a permanent difference exists in the behaviour of platelets from migrainou...

Accumulation of 5-hydroxytryptamine leads to dysfunction of adrenergic nerves in canine coronary artery following intimal damage in vivo.

Previous in vitro studies have demonstrated that coronary artery adrenergic nerves are a principal site of accumulation of 5-hydroxytryptamine released from aggregating platelets. The purpose of this study was to determine whether 5-hydroxytryptamine is accumulated by adrenergic nerves at sites of endothelial damage and platelet aggregation in vivo. Coronary artery 5-hydroxytryptamine content a...

Effects of reserpine and chlorpromazine on 5-hydroxytryptamine uptake of platelets from migrainous and control subjects.

Platelet aggregation experiments confirm differences between the 5-hydroxytryptamine (5-HT) receptors of migrainous and control platelets. However, no differences have been found in 5-HT uptake, in the presence of reserpine or chlorpromazine, into migrainous and control platelets. The results support the hypothesis that receptors for 5-HT induced platelet aggregation provide a model for vascula...

Effect of serotonin on platelet function in cocaine exposed blood

5-hydroxytryptamine (5-HT) reuptake inhibitors counteract the pro-thrombotic effect of elevated plasma 5-HT by down-regulating the 5-HT uptake rates of platelets. Cocaine also down-regulates the platelet 5-HT uptake rates but in contrast, the platelets of cocaine-injected mice show a much higher aggregation rate than the platelets of control mice. To examine the involvement of plasma 5-HT in co...
